Overview

This refers to an intensive induction chemotherapy regimen, commonly known as 'D/IA' or a variation of the '3+7' protocol, which is being evaluated by the Institute of Hematology & Blood Diseases Hospital in China for the treatment of acute myeloid leukemia (AML). The regimen consists of an anthracycline—either daunorubicin or idarubicin—combined with the antimetabolite cytarabine. Daunorubicin and idarubicin are small molecule anthracycline antibiotics that function by intercalating into DNA and inhibiting topoisomerase II, thereby preventing DNA replication and transcription. Cytarabine is a pyrimidine analog that inhibits DNA polymerase, further interfering with DNA synthesis in rapidly dividing cancer cells. In the context of a Phase 3 randomized trial (NCT05030155), this combination serves as the standard-of-care intensive induction therapy arm compared against the combination of venetoclax and azacitidine in elderly patients (aged 55-75) with newly diagnosed AML who are suitable for intensive chemotherapy.
