Overview

Dauricine is a bisbenzylisoquinoline alkaloid isolated primarily from the roots and rhizomes of Menispermum dauricum (Asiatic Moonseed), a plant used in traditional Chinese medicine. It exhibits multiple pharmacological activities including anti-cancer (notably against colon cancer and lung adenocarcinoma), neuroprotective, anti-inflammatory, anti-arrhythmic and anti-diabetic effects. Mechanistically, dauricine acts as an antagonist of thromboxane A2 receptor (TBXA2R), a blocker of voltage-gated calcium channels (VDCCs), and an antagonist of vascular endothelial growth factor receptor 3 (VEGFR3). It also inhibits cell proliferation and invasion by suppressing NF-kappaB activation and modulates apoptosis via mitochondrial pathways. Despite promising preclinical results for various indications such as cancer and neurodegenerative diseases like Alzheimer's disease, its clinical development has been limited due to issues like rapid metabolism and potential pulmonary toxicity[3][5][8][10].
