Overview

Davalintide is a second-generation, 32-amino acid peptide analogue of human amylin developed by Amylin Pharmaceuticals for the treatment of obesity. It acts as an amylin mimetic with enhanced pharmacological properties compared to native amylin. Davalintide binds with high affinity to amylin receptors (as well as calcitonin and calcitonin gene-related peptide receptors), resulting in slowed gastric emptying, inhibition of digestive secretions (gastric acid, pancreatic enzymes, bile), reduced glucagon secretion, and increased satiety. These actions contribute to antihyperglycemic effects and weight loss through decreased food intake and fat-specific weight reduction while sparing lean mass. The drug was investigated in clinical trials for obesity but development was discontinued around 2010[2][3][4][5].
