Dazdotuftide is a novel, synthetic small molecule drug developed as a multitarget anti-inflammatory and immunomodulatory agent for ocular diseases. It is a conjugate of tuftsin and phosphorylcholine, designed to provide potent steroid-like efficacy in controlling autoimmune and inflammatory eye conditions without the adverse effects associated with corticosteroids, such as elevated intraocular pressure (IOP) or glaucoma. Dazdotuftide acts via a dual-target mechanism involving modulation of neuropilin 1 (NRP1), toll-like receptors (TLR), and angiotensin-converting enzyme 2 (ACE2), disrupting key inflammatory pathways implicated in diseases like noninfectious anterior uveitis, diabetic retinopathy, diabetic macular edema, and age-related macular degeneration. The drug has demonstrated favorable safety and efficacy profiles in phase 3 clinical trials for noninfectious anterior uveitis[1][6][8][9][10].
