Overview

Dazodalibep is an investigational recombinant fusion protein designed as a CD40 ligand (CD40L, also known as CD154) antagonist. It blocks the interaction between T cells and CD40-expressing B cells, thereby disrupting the overactivation of the CD40-CD40L co-stimulatory pathway that is implicated in several autoimmune diseases. Dazodalibep is being developed primarily for Sjögren's syndrome, where it has shown efficacy in reducing systemic disease activity and improving symptoms such as dryness and fatigue in clinical trials. The drug was originally developed by MedImmune and Viela Bio, later acquired by Horizon Therapeutics, with current development led by Amgen[1][2][5].
