Overview

**Dazostinag + pembrolizumab** is an investigational combination therapy being researched primarily for advanced or metastatic solid tumors, including squamous cell carcinoma of the head and neck and colorectal cancer. **Dazostinag** (TAK-676) is a small molecule agonist of the stimulator of interferon genes (**STING**) pathway, designed to promote innate and adaptive anti-tumor immune responses. **Pembrolizumab** is a monoclonal antibody targeting the programmed cell death 1 (**PD-1**) receptor, functioning as an immune checkpoint inhibitor to restore and enhance T-cell mediated anti-tumor activity. The combination aims to potentiate anti-tumor immune effects by both directly stimulating innate immunity (via STING activation) and releasing adaptive immune inhibition (via PD-1 blockade). The combination is currently in early-stage clinical trials for solid tumors, including head and neck squamous cell carcinoma (SCCHN) and metastatic colorectal cancer (MSI-H/dMMR and MSS/pMMR subtypes)[1][3][5][7][9].
