Drug intelligence / Profile preview
DB-020 is a novel injectable formulation of sodium thiosulfate (STS) pentahydrate in sodium hyaluronate, designed for intratympanic (IT) administration to the middle ear. Its primary purpose is to prevent or reduce cisplatin-induced ototoxicity (hearing loss) in patients undergoing cisplatin chemotherapy for various cancers. The mechanism of action involves local delivery of high concentrations of thiosulfate to the cochlea, where it acts as a thiol-containing reducing agent that can inactivate cisplatin through covalent binding, thereby protecting inner ear structures from damage without compromising systemic anti-tumor efficacy. DB-020 has demonstrated safety and tolerability in phase 1 clinical trials and shows promise for partial or complete protection against hearing loss compared to placebo[1][2][3][6][7].
