DB-1310 is a novel antibody-drug conjugate (ADC) designed for the treatment of advanced or metastatic solid tumors, particularly those expressing human epidermal growth factor receptor 3 (HER3/ErbB3). It consists of a humanized IgG1 monoclonal antibody targeting HER3, covalently linked via a cleavable peptide linker to a proprietary DNA topoisomerase I inhibitor payload. The ADC binds to HER3 on tumor cells, is internalized, and releases the cytotoxic payload intracellularly to induce tumor cell death. Preclinical studies have shown potent antitumor activity in various HER3-positive cancer models including breast, lung, colon, and prostate cancers. Early clinical trials indicate promising efficacy in heavily pretreated patients with advanced solid tumors—especially EGFR-mutant non-small cell lung cancer (NSCLC)—with manageable side effects primarily consisting of low blood counts and nausea[2][5][6][8][10].
