DB-1314 is a novel antibody-drug conjugate (ADC) targeting delta-like ligand 3 (DLL3), which is highly expressed on the surface of small cell lung cancer (SCLC) cells and minimally in normal tissues. DB-1314 is composed of a humanized anti-DLL3 monoclonal antibody (DB131401) conjugated to eight molecules of P1021, a DNA topoisomerase I inhibitor payload. It binds DLL3 with high affinity and specificity, is internalized into DLL3-positive cells, and upon internalization, releases P1021 to induce DNA damage and cytotoxicity. The mechanism of action involves not only direct DNA damage but also bystander effects and antibody-dependent cell-mediated cytotoxicity (ADCC). DB-1314 demonstrates potent and durable antitumor activity in preclinical in vitro and in vivo SCLC models and shows a favorable safety profile in animal studies, supporting its further clinical evaluation for DLL3-positive SCLC[1][3][4][7].
