DB-1325 is a novel antibody-drug conjugate (ADC) targeting B cell maturation antigen (BCMA), developed by Duality Biologics for the treatment of multiple myeloma. It consists of the monoclonal antibody belantamab conjugated via a cleavable linker to PDUP5, a proprietary "Duality Unique Payload" (DUP) that acts as an mRNA translation inhibitor. This mechanism of action distinguishes it from existing BCMA-targeting ADCs like belantamab mafodotin, which typically utilize microtubule inhibitors. In preclinical studies, DB-1325 demonstrated potent in vitro anti-proliferative activity across various multiple myeloma cell lines and superior in vivo anti-tumor efficacy compared to belantamab mafodotin in xenograft models. Preliminary toxicology assessments in non-human primates have indicated that the drug is well-tolerated with a favorable toxicokinetic profile.
