**DB-1326** is a preclinical, dual-payload antibody-drug conjugate developed by DualityBio for epithelial solid tumors expressing tumor-associated mucin-1. It uses a fully human IgG1 antibody that selectively recognizes the cancer-associated, aberrantly glycosylated TA-MUC1 glycoform and delivers two cytotoxic payload classes: a DNA topoisomerase I inhibitor and a novel ecteinascidin derivative. In preclinical cell-line-derived and patient-derived xenograft models, DB-1326 showed target-dependent internalization, in-vitro cytotoxicity, dose-dependent tumor growth inhibition, and superior antitumor activity relative to a single-payload comparator ADC.
