DB-1329 is a novel antibody-drug conjugate (ADC) developed by DualityBio that targets CUB Domain-Containing Protein 1 (CDCP1), a cell surface protein highly expressed in various solid malignancies, including lung, pancreatic, ovarian, and colorectal cancers. The ADC comprises a high-affinity, fully human anti-CDCP1 monoclonal antibody conjugated to a potent camptothecin-based topoisomerase 1 (TOP1) inhibitor payload via a protease-cleavable linker. Upon binding to CDCP1 on the tumor cell surface, DB-1329 is rapidly internalized, releasing the cytotoxic payload to induce DNA strand breaks and cell death. Preclinical studies have demonstrated significant anti-tumor efficacy and a manageable safety profile, supporting its clinical development for CDCP1-positive solid tumors.
