Drug intelligence / Profile preview
DB-727 is a preclinical-stage, bifunctional fusion protein consisting of a bivalent anti-programmed cell death protein 1 (PD-1) antibody fused to a modified interleukin-2 (IL-2) cytokine. Developed by TrueSimple Bio, DB-727 is designed to target and activate tumor-infiltrating PD-1-positive T cells. The molecule utilizes an IL-2 receptor subunit alpha (IL-2Rα)-biased modification strategy, retaining high affinity for IL-2Rα to selectively stimulate effector T cells while minimizing systemic toxicity. By combining PD-1 checkpoint blockade with localized IL-2 receptor agonism, DB-727 aims to enhance anti-tumor immune responses in solid tumors.
