dbDNA (doggybone DNA) is a linear, double-stranded, covalently closed DNA vector developed by the UK-based biotechnology company Touchlight. It is produced via a cell-free enzymatic manufacturing process, distinguishing it from traditional plasmid DNA which requires bacterial fermentation. This process creates a minimal genetic construct that excludes bacterial sequences such as origins of replication and antibiotic resistance genes, which may enhance safety and reduce the risk of immune responses to bacterial DNA. dbDNA is utilized as a versatile platform for the production of mRNA and viral vectors (including AAV and lentivirus) and is being developed as a direct active pharmaceutical ingredient for DNA vaccines and gene therapies, with clinical applications in oncology.
