Overview

DBP117 is a small molecule, deuterated analog of the (S)-enantiomer of pioglitazone, developed by DeuterRx. Pioglitazone is a thiazolidinedione (TZD) that typically exists as a racemic mixture of (R) and (S) enantiomers which rapidly interconvert in the body. DBP117 utilizes deuterium substitution at the chiral center to stabilize the (S)-enantiomer, which is the primary driver of peroxisome proliferator-activated receptor gamma (PPARγ) activation. By stabilizing this specific enantiomer, DBP117 is intended to provide potent insulin-sensitizing effects for the treatment of type 2 diabetes mellitus. While the (R)-enantiomer (DRX-065) is being pursued for NASH due to its non-PPARγ mediated effects, DBP117 focuses on the classical PPARγ-mediated glucose-lowering pathway.
