Overview

DBXO-1 is a preclinical-stage bispecific T-cell engager (TCE) designed to target multiple cancer-specific peptide-MHC (pMHC) complexes presented by HLA-A*02:01. Developed by Debiopharm, the therapeutic utilizes a single binder engineered to recognize a defined set of intracellular protein-derived peptides, addressing the challenges of low copy number and tumor heterogeneity typically associated with single pMHC targets. The molecule is formatted as an Fc-containing bispecific antibody that recruits T cells via a CD3-binding domain to induce potent, dose-dependent cytotoxicity. Preclinical characterization has demonstrated high specificity and functional activity against solid tumor cell lines, with a first-in-human clinical trial anticipated to begin in 2027.
