Overview

DC-15442 is an orally bioavailable, potent, and selective small molecule inhibitor of Signal Transducer and Activator of Transcription 6 (STAT6), developed by DeepCure using artificial intelligence and advanced physics simulations. It is designed to inhibit the IL-4/IL-13 signaling pathway, which is a central driver of Type 2 inflammation. Unlike STAT6 degraders, DC-15442 acts as a direct inhibitor of the protein, specifically targeting phosphorylated STAT6 (pSTAT6). It is being developed as an oral alternative to injectable biologics, such as dupilumab, for the treatment of chronic inflammatory conditions including asthma, atopic dermatitis, and chronic obstructive pulmonary disease (COPD). Preclinical studies have demonstrated nearly 100% inhibition of pSTAT6 and efficacy comparable to dupilumab across multiple disease models with high selectivity and tolerability.
