Overview

DC-activated donor lymphocyte infusion (DC/ADL) is an experimental adoptive cellular immunotherapy developed by the M.D. Anderson Cancer Center. This personalized therapy is designed for patients with relapsed acute myeloid leukemia (AML) or chronic myeloid leukemia (CML) in blast crisis following an allogeneic stem cell transplantation. The therapeutic process involves the ex vivo generation of dendritic cells from the patient's own leukemic blasts, which are then used to prime and activate allogeneic donor lymphocytes. These activated donor T-cells are infused back into the patient to enhance the graft-versus-leukemia (GVL) effect. By specifically targeting leukemia-associated antigens presented by the dendritic cells, the therapy aims to stimulate a potent anti-leukemic immune response while attempting to minimize the risk of systemic graft-versus-host disease (GVHD).
