DC-Ad-GMCAIX is an autologous dendritic cell-based cancer vaccine designed to treat metastatic renal cell carcinoma (RCC). The therapy involves harvesting a patient's own dendritic cells and transducing them with an adenoviral vector that encodes two key components: the tumor-associated antigen Carbonic Anhydrase IX (CAIX) and the immunostimulatory cytokine Granulocyte-Macrophage Colony-Stimulating Factor (GM-CSF). CAIX is a cell surface protein overexpressed in over 90% of clear cell renal cell carcinomas, making it a specific target for immunotherapy. GM-CSF serves as a potent adjuvant to promote the maturation and activation of the dendritic cells, enhancing their ability to present CAIX antigens to T cells. Once re-infused into the patient, these engineered dendritic cells stimulate a robust, CAIX-specific cytotoxic T-lymphocyte (CTL) response aimed at destroying tumor cells.
