DC-cholesterol is a synthetic, **cationic cholesterol derivative** primarily used as a gene and drug delivery vehicle in research and experimental therapeutics. It enhances the formation of stable cationic liposomes, which are routinely combined with helper lipids such as DOPE (1,2-di-(9Z-octadecenoyl)-sn-glycero-3-phosphoethanolamine) to form liposomes capable of encapsulating and delivering nucleic acids (DNA, siRNA, mRNA) and some small molecule drugs into mammalian cells. The positively charged ammonium group at the 3-position allows for electrostatic interaction with negatively charged nucleic acids, facilitating cellular uptake via pathways such as macropinocytosis and clathrin-mediated endocytosis. DC-cholesterol-based liposome formulations are noted for efficient gene transfection and relatively low cytotoxicity compared to other cationic lipids[3][4][8][9].
