**DC-shA20-FAP-TRP2** is an experimental genetically modified dendritic-cell cancer vaccine developed in an academic preclinical program. The vaccine incorporates an A20-specific short hairpin RNA to suppress the negative regulator A20 in dendritic cells and expresses fibroblast activation protein and tyrosinase-related protein 2 antigens, aiming to generate cytotoxic T-cell responses against both fibroblast activation protein-positive cancer-associated fibroblasts and tyrosinase-related protein 2-positive melanoma cells. In the syngeneic B16 melanoma mouse model, it increased fibroblast activation protein- and tyrosinase-related protein 2-specific T-cell responses, tumor CD8-positive T-cell infiltration, antigen spreading, and antitumor activity relative to monovalent or control-shRNA vaccine comparators.
