DC-VACCINE IRST-IRCCS is an autologous dendritic cell-based immunotherapy developed by the Istituto Romagnolo per lo Studio dei Tumori (IRST). The vaccine is produced by isolating monocytes from a patient's peripheral blood, differentiating them into mature dendritic cells ex vivo, and loading them with tumor-associated antigens, typically derived from autologous tumor lysate. Upon re-administration, these primed dendritic cells migrate to lymph nodes and present the antigens to T-lymphocytes, inducing a specific cytotoxic immune response against the patient's tumor. This approach is primarily being investigated for refractory metastatic colorectal cancer (mCRC) with mismatch repair-proficient (pMMR) or microsatellite-stable (MSS) status, as well as glioblastoma, often in combination with immune checkpoint inhibitors to overcome the immunosuppressive tumor microenvironment.
