Overview

DC561043 is a novel small molecule antidepressant developed by the Shanghai Institute of Materia Medica (SIMM), Chinese Academy of Sciences. It functions as a triple reuptake inhibitor (TRI), simultaneously targeting the serotonin transporter (SERT), norepinephrine transporter (NET), and dopamine transporter (DAT). By inhibiting the reuptake of these three key neurotransmitters, DC561043 increases their synaptic concentrations, which is hypothesized to provide a more rapid onset of action and superior efficacy compared to traditional selective serotonin reuptake inhibitors (SSRIs) or dual reuptake inhibitors. Preclinical studies demonstrated significant antidepressant-like activity in various animal models. The drug received clinical trial approval from the China National Medical Products Administration (NMPA) in late 2021 and is currently being evaluated in early-phase clinical trials for the treatment of major depressive disorder.
