Overview

dCas9-DNMT is an epigenome-editing tool consisting of a catalytically inactive "dead" Cas9 (dCas9) protein fused to a DNA methyltransferase (DNMT) domain. Unlike traditional CRISPR-Cas9 which induces double-strand breaks, dCas9-DNMT is guided by a specific guide RNA (gRNA) to a target genomic locus where it catalyzes the addition of methyl groups to CpG sites. This targeted DNA hypermethylation typically leads to transcriptional silencing of the target gene. In research contexts, such as studies on Crohn's disease, it is used to model the effects of gene hypermethylation (e.g., the *SP140* gene) on immune cell function and cytokine production (e.g., TNFα, IL-6).
