DCB-AB21 is a humanized monoclonal antibody that targets the colony-stimulating factor 1 receptor (CSF-1R). By specifically binding to CSF-1R, it inhibits downstream signaling and blocks cell proliferation induced by the ligands CSF-1 and IL-34. The therapeutic strategy focuses on modulating the tumor microenvironment by targeting tumor-associated macrophages (TAMs), which are known to contribute to immune suppression. Preclinical data indicates that DCB-AB21 exhibits moderate anti-cancer activity as a monotherapy and can significantly enhance the efficacy of anti-PD-1 checkpoint inhibitors. The antibody was originally screened from immunized mouse hybridoma clones and underwent humanization engineering to optimize its affinity and anti-proliferative properties.
