Overview

DCB-BO1301 + dacarbazine is an investigational combination therapy comprising a humanized monoclonal antibody targeting CD47 and the cytotoxic alkylating agent dacarbazine. DCB-BO1301 functions by binding to CD47, a cell surface protein often overexpressed on cancer cells that serves as a "don't eat me" signal by interacting with SIRPα on macrophages. By blocking this interaction, DCB-BO1301 promotes the phagocytosis of tumor cells by the innate immune system. Dacarbazine is a prodrug that undergoes metabolic activation to form a methylating agent, which alkylates DNA at the O6 and N7 positions of guanine, leading to DNA strand breaks and apoptosis. The combination is primarily being explored for the treatment of advanced solid tumors, such as metastatic melanoma, aiming to synergize the immunomodulatory effects of CD47 blockade with the direct cytotoxic activity of chemotherapy.
