Overview

DCB-HDG2-57 is a small molecule dual mTOR-PI3K inhibitor that also possesses Hedgehog (Hh) signaling pathway antagonist activity. Developed by the Development Center for Biotechnology (DCB) in Taiwan, the compound is designed to address resistance in Hh-dependent tumors by simultaneously inhibiting the PI3K/AKT/mTOR pathway, which is often hyperactivated in cancers and linked to drug resistance. DCB-HDG2-57 demonstrates potent biochemical inhibition of PI3Kα and mTOR, as well as cellular inhibition of Smoothened (Smo), including the Smo D473H mutant. Preclinical evidence shows its ability to reduce cancer stem cell-like populations and inhibit tumor growth in colon and pancreatic cancer models.
