Overview

DCC-3084 is an orally bioavailable, central nervous system (CNS)-penetrant, small molecule pan-RAF inhibitor developed by Deciphera Pharmaceuticals. It is designed to broadly inhibit Class I, II, and III BRAF mutations, BRAF fusions, and BRAF/CRAF heterodimers. The drug acts as a switch-control inhibitor of the serine/threonine protein kinase Raf family and demonstrates potent inhibition of MAPK pathway signaling in preclinical models. DCC-3084 has shown antitumor activity in various RAS/RAF mutant cancers—including those with KRAS or NRAS mutations—and exhibits favorable pharmaceutical properties compared to earlier pan-RAF inhibitors. Its CNS penetration suggests potential utility for brain metastases or primary brain tumors. Currently, it is being evaluated in Phase 1/2 clinical trials for advanced solid tumors driven by MAPK pathway alterations such as non-small cell lung cancer (NSCLC), pancreatic ductal adenocarcinoma (PDAC), melanoma, prostate cancer (including castration-resistant forms), and other RAF/RAS/NF1-mutated malignancies.
