DCLK1 CAR-T cells are an experimental chimeric antigen receptor (CAR) T-cell therapy designed to target Doublecortin-like kinase 1 (DCLK1), a marker associated with cancer stem cells (CSCs). Developed through a collaboration between COARE Holdings and ProMab Biotechnologies, these CAR-T cells utilize a humanized monoclonal antibody clone (CBT-15 #8) that recognizes a specific extracellular C-terminal epitope on human DCLK1 isoforms 2 and 4. DCLK1 is highly expressed in multiple myeloma cells and various solid tumors, such as colon and pancreatic cancer, but is relatively restricted in normal tissues, making it a potential target for eliminating the cell populations responsible for tumor initiation, progression, and therapy resistance. Preclinical studies have demonstrated significant cytotoxicity and interferon-gamma secretion against multiple myeloma cell lines.
