DCLK1 siRNA is an experimental small interfering RNA (siRNA) designed to silence the expression of the Doublecortin-Like Kinase 1 (DCLK1) gene through RNA interference. DCLK1 is a serine/threonine kinase that serves as a marker for tumor stem cells and is overexpressed in various malignancies, including colorectal, pancreatic, and renal cell cancers, as well as in fibrotic conditions like idiopathic pulmonary fibrosis (IPF). In preclinical models, knockdown of DCLK1 using siRNA has been shown to inhibit epithelial-mesenchymal transition (EMT), reduce cancer stem cell markers, decrease tumor cell invasion and migration, and sensitize cells to chemotherapy and radiation. In the context of pulmonary fibrosis, DCLK1 siRNA attenuates fibroblast activation, myofibroblast transformation, and extracellular matrix production. It is primarily used as a research tool in academic laboratories and has been evaluated in various delivery systems, such as liposomes and PLGA nanoparticles.
