DCTRS4-406 (also referred to as CCT303-406) is an autologous chimeric antigen receptor T-cell (CAR-T) therapy developed by Exuma Biotechnology for the treatment of HER2-overexpressing solid tumors. The therapy incorporates Exuma's proprietary Tumor Metabolism Regulated (TMR) technology, also known as Tumor Microenvironment Restricted technology. This platform is designed to conditionally activate the CAR-T cells only within the unique metabolic or chemical conditions of the tumor microenvironment, thereby minimizing the risk of on-target, off-tumor toxicity against healthy tissues that express low levels of the HER2 protein. DCTRS4-406 has completed a Phase 1 investigator-initiated clinical trial that demonstrated promising safety and preliminary efficacy in patients with HER2-positive cancers, including gastroesophageal, breast, and colorectal malignancies.
