Overview

DD-IL12 is an experimental immunotherapeutic agent developed by Obsidian Therapeutics using its proprietary cytoDRIVE platform. It consists of the cytokine Interleukin-12 (IL-12) fused to a destabilizing domain (DD), which renders the protein unstable and targets it for rapid proteasomal degradation in the basal state. Upon administration of a specific small-molecule stabilizer (ligand), such as the FDA-approved drug trimethoprim (TMP), the DD is stabilized, allowing the IL-12 payload to accumulate and exert its biological effects in a dose-dependent and reversible manner. This technology is primarily designed to "armor" adoptive cell therapies, such as CAR-T cells or tumor-infiltrating lymphocytes (TILs), enabling localized cytokine activity within the tumor microenvironment to enhance anti-tumor efficacy while avoiding the severe systemic toxicities typically associated with constitutive IL-12 expression.
