DD-IL15 (also known as PMI06) is a long-acting recombinant fusion protein designed to harness the immunostimulatory potential of Interleukin-15 (IL-15) for oncology applications. Developed by D&D Pharmatech and its subsidiary Precision Molecular, the molecule consists of human IL-15 fused to the sushi domain of the IL-15 receptor alpha (IL-15Rα), which is further stabilized by a dimerization domain to extend its half-life and increase potency. This configuration mimics the natural "trans-presentation" of IL-15, significantly increasing its stability and biological activity compared to recombinant IL-15 alone. DD-IL15 acts as an agonist for the dimeric IL-15 receptor complex, specifically targeting the IL-2/IL-15 receptor beta (CD122) and common gamma (CD132) subunits. This stimulation leads to the selective expansion and activation of cytotoxic CD8+ T cells and Natural Killer (NK) cells, which are critical for anti-tumor immunity, while minimizing the activation of immunosuppressive regulatory T cells (Tregs). It is currently being investigated in clinical trials for the treatment of advanced solid tumors.
