DDD-028 is a non-opioid small molecule analgesic being developed by Daya CNS for the treatment of various chronic pain conditions, including neuropathic pain, arthritis, and fibromyalgia. Chemically, it is a pentacyclic pyridoindolobenzazepine derivative (also described as a pyridoindole derivative). Its mechanism of action is distinct from traditional analgesics as it does not interact with opioid, cannabinoid, dopamine, or histamine receptors; instead, its anti-hyperalgesic effects are mediated through the nicotinic system, specifically targeting the alpha-7 nicotinic acetylcholine receptor (α7 nAChR). Beyond its analgesic properties, DDD-028 has demonstrated neuroprotective effects in preclinical models, particularly against chemotherapy-induced peripheral neuropathy (CIPN) caused by paclitaxel, by inhibiting glial cell proliferation and preventing peripheral nervous system damage.
