DDOX is a non-antibiotic derivative of the tetracycline antibiotic doxycycline, specifically 4-de(dimethylamino)doxycycline. It belongs to a class of compounds known as Chemically Modified Tetracyclines (CMTs), which are engineered to lack antimicrobial activity while retaining or enhancing other pharmacological properties such as anti-inflammatory, antioxidant, and neuroprotective effects. DDOX has been investigated for its potential to rescue dopaminergic neurons from iron-dependent ferroptosis, a process implicated in the pathogenesis of Parkinson's disease. Its mechanism involves the prevention of intracellular oxidative stress and the maintenance of mitochondrial membrane potential, thereby mitigating lipid peroxidation-mediated cellular damage. Research indicates that DDOX is particularly effective against chronic, low-intensity oxidative insults, suggesting potential therapeutic utility in the long-term management of neurodegenerative conditions.
