DDX11 siRNA is an experimental RNA interference (RNAi) therapeutic agent designed to silence the expression of the DEAD-box helicase 11 (DDX11) gene, also known as ChlR1. DDX11 is a DNA helicase that plays a pivotal role in maintaining genome stability through its involvement in sister chromatid cohesion, DNA replication, and the repair of DNA interstrand cross-links. In various cancers, such as melanoma and osteosarcoma, DDX11 is frequently overexpressed, contributing to tumor cell proliferation, survival, and resistance to DNA-damaging therapies. By utilizing the RNA-induced silencing complex (RISC), DDX11 siRNA guides the sequence-specific cleavage of DDX11 mRNA, resulting in reduced protein levels. This knockdown induces DNA damage, cell cycle arrest, and apoptosis, making it a subject of preclinical investigation as a potential antineoplastic strategy.
