Overview

DE-CTPI (also known as DE-130 or DE-130A in early development) is a C-terminal peptide inhibitor developed by Santen Pharmaceutical for the treatment of glaucoma and ocular hypertension. The drug is a peptide derived from the C-terminus of the RhoA protein, a small GTPase that plays a critical role in regulating the actin cytoskeleton and cell morphology. In the eye, RhoA activity is involved in the regulation of aqueous humor outflow through the trabecular meshwork. By inhibiting RhoA, DE-CTPI promotes relaxation of the trabecular meshwork and increases the facility of aqueous humor outflow, thereby lowering intraocular pressure (IOP). The asset reached Phase 1/2 clinical trials in Japan; however, development was subsequently discontinued. Note that the code DE-130A was later reused by Santen for a different product, a cationic emulsion formulation of latanoprost (Catioprost).
