Overview

Death receptor 5-targeted chimeric antigen receptor T cells (anti-DR5 CAR-T cells) is an investigational cell therapy being developed by the Shenzhen Hospital of the Chinese Academy of Sciences. The therapy consists of autologous or donor-derived T cells engineered to express a chimeric antigen receptor (CAR) targeting Death Receptor 5 (DR5, also known as TRAIL-R2 or CD262), a member of the tumor necrosis factor receptor superfamily. DR5 is a pro-apoptotic receptor that is upregulated in many solid tumors, including melanoma, breast, lung, and colon cancers, as well as on myeloid-derived suppressor cells (MDSCs). By targeting DR5, these CAR-T cells aim to induce direct tumor cell lysis and potentially modulate the immunosuppressive tumor microenvironment. The therapy is currently being evaluated in an open-label, single-center, single-arm clinical study for the treatment of advanced solid tumors.
