DEBIO 0123 + carboplatin + etoposide is a combination regimen consisting of DEBIO 0123, a brain-penetrant, highly selective WEE1 kinase inhibitor, and the chemotherapeutic agents carboplatin and etoposide. DEBIO 0123 inhibits WEE1, a key regulator of the G2/M and S phase cell cycle checkpoints, which facilitates DNA repair before cells proceed to mitosis. Inhibition of WEE1 abrogates DNA damage checkpoints, causing cells to undergo mitosis without repairing DNA, leading to mitotic catastrophe and apoptosis in cancer cells. Combining DEBIO 0123 with DNA-damaging agents like carboplatin (a platinum-based DNA crosslinker) and etoposide (a topoisomerase II inhibitor) results in enhanced DNA damage and cell death in tumor cells. This combination is currently being developed and has been studied in early phase clinical trials for advanced and/or difficult-to-treat solid tumors, with particular interest in platinum-resistant ovarian cancer[1][5].
