Debio 1453

Debio 1453 is a first-in-class small molecule antibiotic developed by Debiopharm for the treatment of Neisseria gonorrhoeae infections, including drug-resistant and multidrug-resistant strains. It acts as a potent inhibitor of FabI (enoyl-acyl carrier protein reductase), an essential enzyme in the bacterial fatty acid synthesis (FASII) pathway. By inhibiting FabI, Debio 1453 disrupts fatty acid biosynthesis required for bacterial growth and survival, resulting in rapid bactericidal activity against N. gonorrhoeae. The compound demonstrates high efficacy with a favorable resistance profile and no cross-resistance to existing antibiotics, while sparing healthy microbiota due to its targeted spectrum. It is being developed for both oral and intramuscular administration and has also shown promising preclinical activity against Chlamydia trachomatis[1][2][4][5][6].
