Decitabine + autologous DC-CIK cells is a combination chemo-immunotherapy regimen being investigated as a post-remission maintenance treatment for malignant solid tumors. The regimen consists of the hypomethylating agent decitabine followed by the infusion of autologous dendritic cell-cytokine induced killer (DC-CIK) cells. Decitabine, a DNA methyltransferase inhibitor, is administered at low doses to induce epigenetic changes in tumor cells, such as the upregulation of tumor-associated antigens and MHC molecules, which sensitizes the tumor to immune-mediated destruction. DC-CIK cells are a heterogeneous population of immune effector cells, primarily CD3+CD56+ T cells, that are expanded ex vivo with cytokines and pulsed with dendritic cells to enhance their tumor-specific targeting and cytotoxic activity. This combination therapy aims to leverage the epigenetic priming of decitabine to improve the efficacy of adoptive cell transfer in patients with solid tumors.
