The DCHA regimen is a multi-agent combination chemotherapy protocol developed by the Chinese PLA General Hospital for the treatment of acute myeloid leukemia (AML), specifically as post-remission consolidation therapy for patients with the t(8;21) chromosomal translocation. The regimen integrates epigenetic modulators with traditional cytotoxic agents, consisting of decitabine (a hypomethylating agent), chidamide (a selective histone deacetylase inhibitor), homoharringtonine (a plant alkaloid protein synthesis inhibitor), and cytarabine (an antimetabolite). By targeting multiple pathways—including DNA methylation, histone deacetylation, and DNA synthesis—the DCHA regimen aims to eradicate minimal residual disease and reduce relapse rates in t(8;21) AML patients, a subgroup that often carries KIT mutations associated with a higher risk of recurrence despite a generally favorable prognosis.
