Overview
Decitabine + daunorubicin + cytarabine is a combination chemotherapy regimen used for the treatment of **acute myeloid leukemia (AML)**, especially in older patients or those with poor-risk cytogenetics. Each component functions via distinct mechanisms: - **Decitabine** is a hypomethylating agent that inhibits DNA methyltransferase, leading to hypomethylation of DNA and subsequent reactivation of silenced genes involved in cell cycle control and apoptosis, thereby promoting cell death in malignant cells[2][6]. - **Daunorubicin** is an anthracycline antibiotic that intercalates into DNA, inhibits topoisomerase II, and generates free radicals, resulting in DNA damage and inhibition of cell replication[5][7]. - **Cytarabine** is an antimetabolite that inhibits DNA polymerase, interfering with DNA synthesis during the S phase of the cell cycle[5][7]. Clinical trials have investigated the efficacy of this combination in the induction and consolidation phases for AML, and compared it with or sequential addition of decitabine following other regimens[3][4].
