Overview

Decitabine + Hiltonol is an investigational combination therapy consisting of two agents: - Decitabine is a small molecule chemotherapeutic and epigenetic modulator classified as a DNA methyltransferase inhibitor. It induces DNA hypomethylation by incorporating into DNA and inhibiting DNA methyltransferases, leading to reactivation of silenced genes involved in tumor suppression and immune response. Decitabine is approved for myelodysplastic syndromes and has been studied in acute myeloid leukemia and other malignancies[3][4][5][6]. - Hiltonol is the trade name for Poly-ICLC, a synthetic double-stranded RNA complex that acts as an immunostimulant by activating Toll-like receptor 3 (TLR3) on dendritic cells and other immune cells. This activation leads to induction of type I interferons and pro-inflammatory cytokines, enhancing innate immunity against tumors. The combination aims to leverage the epigenetic reprogramming effects of decitabine with the immune-stimulating properties of Hiltonol/Poly-ICLC to enhance anti-tumor immunity. This approach has been explored in early-phase clinical trials for relapsed cancers[1].
