Overview

A combination therapy consisting of decitabine, nivolumab, and tetrahydrouridine. Decitabine depletes DNA methyltransferase 1 (DNMT1), an enzyme often increased in cancer that suppresses neo-antigen expression. Tetrahydrouridine (THU) blocks cytidine deaminase, an enzyme that rapidly breaks down decitabine, thereby increasing the time cancer cells are exposed to decitabine. Nivolumab is an immune checkpoint inhibitor that blocks PD-1, enhancing T-cell immune response against cancer cells. This combination aims to overcome the limitations of using these drugs individually, particularly for solid tumors like non-small cell lung cancer (NSCLC).
