Overview

Decitabine + panitumumab is an investigational combination therapy consisting of decitabine, a hypomethylating agent, and panitumumab, a fully human monoclonal antibody targeting the epidermal growth factor receptor (EGFR). Decitabine acts as a DNA methyltransferase inhibitor, leading to hypomethylation of DNA and potential reactivation of tumor suppressor genes. Panitumumab binds to EGFR on the surface of cancer cells, blocking ligand-induced activation and downstream signaling pathways involved in cell proliferation and survival. This combination has been studied primarily in patients with metastatic colorectal cancer (mCRC) who have wild-type KRAS tumors. The rationale for combining these agents is that epigenetic modulation by decitabine may enhance sensitivity to anti-EGFR therapy in resistant tumors[1][2].
