Overview

Decoglurant is a small molecule drug that acts as a negative allosteric modulator (antagonist) of the metabotropic glutamate receptor 2 (mGlu2) and metabotropic glutamate receptor 3 (mGlu3). It was developed by Roche for adjunctive treatment of major depressive disorder, with the aim to provide antidepressant effects by modulating glutamatergic neurotransmission. Decoglurant advanced to phase II clinical trials but was discontinued due to lack of efficacy in clinical studies. The drug was also investigated for dementia but did not progress beyond early development[1][3][5][7].
