DEDN6526A is a humanized antibody-drug conjugate (ADC) designed to target the endothelin B receptor (ETBR), which is often overexpressed in melanoma and other tumor cells. The drug consists of a monoclonal antibody that specifically binds to ETBR on tumor cells and is linked to the cytotoxic agent monomethyl auristatin E (MMAE). Upon binding to ETBR-expressing cells, DEDN6526A is internalized; proteolytic cleavage then releases MMAE inside the cell. MMAE binds tubulin and inhibits its polymerization, leading to G2/M phase cell cycle arrest and apoptosis of the cancer cell. DEDN6526A was developed primarily for metastatic or unresectable melanoma[1][2][3][4][5].
