def-22B is a humanized monoclonal antibody designed to target Syndecan-1 (SDC1), a cell-surface heparan sulfate proteoglycan that is significantly upregulated in pancreatic ductal adenocarcinoma (PDAC) and other cancers driven by KRAS mutations. SDC1 plays a pivotal role in macropinocytosis, a process where cancer cells engulf extracellular fluids to salvage nutrients, supporting their growth in the nutrient-depleted tumor microenvironment. def-22B works by binding to SDC1, which inhibits this macropinocytotic pathway and induces antibody-dependent cellular cytotoxicity (ADCC) against the tumor cells. Developed by DeFini Therapeutics, def-22B has demonstrated the ability to suppress tumor growth in preclinical models, both as a monotherapy and in combination with standard-of-care chemotherapy, KRAS inhibitors, and immunotherapies.
