DEG6498

DEG6498 is a first-in-class, orally bioavailable small molecule molecular glue degrader developed by Degron Therapeutics. It targets the RNA-binding protein Human antigen R (HuR, also known as ELAVL1), which has been considered "undruggable" by conventional approaches. HuR plays critical roles in cancer progression, inflammatory diseases, and metabolic disorders and is often overexpressed or cytoplasmically enriched in many cancers. DEG6498 induces the interaction between the E3 ubiquitin ligase component cereblon (CRBN) and HuR, leading to targeted degradation of HuR protein via the ubiquitin-proteasome system. The drug is being developed for multiple solid tumors including colorectal cancer, glioblastoma, and hepatocellular carcinoma. As of early 2025, an FDA Investigational New Drug (IND) application has been cleared to initiate Phase I clinical trials in solid tumors[1][2][5][6].
